Comparative carcinogenicity in Swiss mice of N-(4-(5-nitro-2-furyl)-2-thiazolyl)acetamide and structurally related 5-nitrofurans and 4-nitrobenzenes.

Aq4{5-Nitro-2-furyl)-2-thiazolyl]acetamide (NFTA), fed to female Swiss mice at doses of 0.05 and 0.1% of the diet, induced a high incidence of lymphocytic leukemia and a low incidence of forestomach tumors. Several other chemicals structurally related to NFTA were also fed to female Swiss mice to determine structure-activity relationships with respect to organ specificity. 7V-[4-(5-Nitro-2-furyl)-2-thiazolyl]formamide induced urinary bladder carcinomas in 20 of 21 mice and low incidences of leukemia and forestomach tumors. 2,2,2-Trifluoro-/V-[4{5-nitro-2-furyl)-2-thiazolyl]acetamide, 2-amino-4-(5-nitro-2-furyl)thiazole, A^-[5-(5-nitro-2-furyl)l,3,4-thiadiazol-2-yl]acetamide, formic acid 2-[4-{5-nitro-2furyl)-2-thiazolyl] hydrazide, 2-(2,2-dimethylhydrazino)-4(5-nitro-2-furyl)thiazole, and 2-hydrazino-4-(5-nitro-2furyl)thiazole induced high incidences of forestomach tumors (23 of 25, 24 of 27,21 of 22, 12 of 20, 13 of 23, and 10 of 17, respectively). 7V-[4-(5-Nitro-2-furyI)-2-thiazolyl] forma mide, formic acid 2-[4-(5-nitro-2-furyl)-2-thiazolyl] hydrazide, 2-hydrazino-4-(p-nitrophenyl)thiazole, 2-hydrazino-4-(paminophenyl)thiazole, and 2-amino-4-(p-nitrophenyl)thiazole induced low incidences of lymphocytic leukemia (6 of 21, 10 of 20, 5 of 21, 5 of 22, and 5 of 20, respectively) with considerably longer latent periods than was the case with NFTA.
